SOP For Cleaning of Manufacturing Equipment


1.0 PURPOSE
To describe a standardized method for cleaning of manufacturing equipment to ensure product quality, prevent cross-contamination, maintain equipment performance, and comply with Good Manufacturing Practices (GMP). This SOP ensures that all production equipment is cleaned, inspected, documented, and released for use in a consistent and controlled manner.
2.0 SCOPE
This SOP is applicable to all manufacturing equipment used in the Production Department, including mixers, granulators, fluid bed dryers, tablet compression machines, coating machines, capsule filling machines, and ancillary equipment. It covers cleaning activities performed after product changeover, batch completion, maintenance work, and scheduled cleaning.
3.0 RESPONSIBILITY
3.1 Production Operator is responsible for performing equipment cleaning as per this SOP and completing all required cleaning records.
3.2 Production Supervisor is responsible for supervising cleaning activities, verifying documentation, and ensuring GMP compliance.
3.3 Quality Assurance (QA) is responsible for verifying cleaning effectiveness, reviewing cleaning records, and approving equipment for production use.
3.4 Engineering Department is responsible for repairing equipment defects and maintaining manufacturing equipment in good working condition.
4.0 ACCOUNTABILITY
4.1 Production Manager is accountable for ensuring the effective implementation of this SOP within the Production Department.
4.2 Head of Quality Assurance (QA) is accountable for monitoring compliance with this SOP and ensuring GMP requirements are met.
4.3 All personnel involved are accountable for following this SOP and maintaining accurate documentation.

6.0 TRAINING
6.1 All personnel shall receive training on this SOP before performing equipment cleaning activities.
6.2 Training shall cover GMP requirements, equipment cleaning procedures, safe chemical handling, personal hygiene, and documentation practices.
6.3 Retraining shall be conducted after SOP revisions, process changes, significant deviations, or as part of annual GMP refresher training.
7.0 PROCEDURE
7.1 Preparation
7.1.1 Verify the equipment is switched off and disconnected from the power supply, if applicable.
7.1.2 Confirm that the equipment is labeled “TO BE CLEANED” before starting the cleaning process.
7.1.3 Wear the required Personal Protective Equipment (PPE).
7.1.4 Replace the “TO BE CLEANED” label with “UNDER CLEANING” before commencing cleaning.
7.1.5 Assemble approved cleaning agents, tools, and materials.
7.2 Pre-Cleaning
7.2.1 Remove all product residues and waste materials from the equipment.
7.2.2 Dismantle removable parts as per the equipment manual.
7.3 Cleaning
7.3.1 Clean all product-contact surfaces using the approved cleaning agent.
7.3.2 Scrub difficult-to-clean areas with approved brushes.
7.3.3 Rinse thoroughly with purified water until no detergent residue remains.
7.4 Drying
7.4.1 Dry all equipment parts using lint-free cloths or filtered compressed air.
7.4.2 Ensure the equipment is completely dry before reassembly.
7.5 Inspection
7.5.1 Visually inspect the equipment for cleanliness and absence of residues.
7.5.2 Report any defects or abnormalities to the Production Supervisor.
7.6 Reassembly
7.6.1 Reassemble the equipment according to the manufacturer’s instructions.
7.6.2 Affix the “CLEANED” status label after successful cleaning.
7.7 Documentation
7.7.1 Record all cleaning activities in the equipment cleaning logbook.
7.7.2 Obtain the required operator, supervisor, and QA signatures before releasing the equipment for production.
8.0 PRECAUTIONS
8.1 Wear appropriate Personal Protective Equipment (PPE) during cleaning.
8.2 Use only approved cleaning agents and equipment.
8.3 Prevent cross-contamination during cleaning operations.
8.4 Ensure equipment is completely dry before use.
8.5 Report any equipment damage or abnormalities immediately.
