1. PURPOSE
To describes the steps for operating the Karl Fischer (KF) apparatus to determine the moisture content in a sample.
2. SCOPE
This SOP is applicable for all personnel operating the Karl Fischer apparatus in the laboratory in pharmaceuticals.

6. PRECAUTIONS
6.1 Use appropriate Personal Protective Equipment (PPE), including gloves, lab coat, and safety goggles.
6.2 Handle Karl Fischer reagent with care, as it may contain toxic chemicals like iodine and sulfur dioxide.
6.3 Work in a well-ventilated area or under a fume hood to minimize inhalation risks.
6.4 Ensure all reagents and samples are disposed of according to lab guidelines.
7. PROCEDURE
7.1 Preparation of Equipment
7.1.1 Verify that the titration cell, electrodes, and other parts of the apparatus are clean and free of contaminants.
7.1.2 If using a titrant solution that has not been pre-prepared, ensure the Karl Fischer reagent is correctly mixed according to the manufacturer’s instructions.
7.2 Sample Preparation
7.2.1 Weigh an appropriate amount of sample using an analytical balance. The amount depends on the expected moisture content and the type of sample being tested.
7.2.2 For solid or semi-solid samples, dissolve the sample in an appropriate solvent or prepare the sample as required (e.g., homogenizing or pulverizing).
7.2.3 Insert the weighed sample into the titration cell. For solid samples, ensure thorough dispersion in the reagent using an appropriate method.
7.2.4 Inject the liquid sample straight into the device if you’re using one.
7.3 Running the Titration
7.3.1 Switch on the Karl Fischer apparatus and allow the system to warm up.
7.3.2 Ensure that the titration cell is filled with the Karl Fischer reagent and connected to the sample chamber.
7.3.3 Set the machine as appropriate for operation.
7.3.4 Begin the titration cycle. The apparatus will automatically add Karl Fischer reagent to the sample.
7.3.5 Monitor the titration process on the apparatus display. The instrument should automatically detect the end point, where the reaction is complete.
7.3.6 At the end of the titration, the apparatus will display the moisture content in percentage or mg of water in the sample. Record the value.
7.4 Cleaning
7.4.1 After each titration, thoroughly clean the titration cell, electrodes, and other components with the recommended solvent (often alcohol or water) to prevent contamination.
7.4.2 Dispose of Samples and Reagents: Follow lab guidelines for proper disposal of waste materials, including Karl Fischer reagent and samples..
7.5 Data Recording and Reporting
7.5.1 Ensure that all titration results, sample details (e.g., type, weight), and any other relevant observations are recorded in the lab notebook or electronic record system.
7.5.2 Generate a report summarizing the moisture content, including any procedural notes, calibration information, and potential sources of error.
7.6 Maintenance
7.6.1 Perform regular maintenance of the Karl Fischer apparatus, including checking for leaks in the titration cell and cleaning the electrodes.
